Vermeule is the daughter of classicist Emily Vermeule and former Museum of Fine Arts curator Cornelius Clarkson Vermeule III.

Her brother, Adrian Vermeule, is a professor at Harvard Law School.

Her wife is Terry Castle, also a professor of English at Stanford.
